Systemic Lupus Erythematosus

Also T cells obtained from females having achieved menopause did not show any significant sensitivity towards intracellular Calcineurin (Rider et al, 1998). This experiment was successful in proving the fact that it is female hormone estrogen that makes female more susceptible to acquire autoimmune disease such as SLE.
Identification of the auto-antibodies involved in systemic lupus is very critical in understanding the way one's own antibody works against one's own cells (Lim et al, 2002). Auto-antibodies associated with Systemic Lupus Erythematosus remained unidentified until early 1990s, analyzing the nature and molecular working of the such auto-antibodies after its identification was still a challenge for the scientific community. Dr. Yoon Lim with team, at The Seoul National University, college of Medicine took the challenge. Sixty eight patients with age range of 26.5years to 55.5years while 78 normal people were used as controls. Samples of ploy (A)+ were collected from human kidney, human testis and human lung cancer cell. After obtaining the samples cDNA expression libraries were constructed. Each library was consisted approximately 2X106 primary recombinants on average and 5 x 105 of them were used for immunoscreening, CDNA expression libraries were screened with sera of ten patients of SLE. Once the immuneoscreenoing was done, the DNA of the sera-raective clones was purified (Lim et al, 2002). The clone sequencing was performed by using genetic analyzer automated sequencer. Serum of patients was used at a dilution of 1:250. The tested clones were easily distinguished from the control clones ,thus could be said to have auto-antibodies against the cell nuclei and cytoplasm (Lim et al, 2002).
The experiment performed by Lim not only showed the presence of the auto-anitbodies against the cell nuclei and cytoplasm in the patients but also identified the exact nature and genetic location of such auto-antibodies (Lim et al, 2002).
