Indirectly-elicited twitches were set up and these results recorded. Then the elicited twitches were stopped and 0.3ml of 1*10-4M Acetylcholine (concentration was final bath concentration, as are all concentrations discussed in this report) and allowed to remain in contact with the tissue for approximately 30s before the organ bath was washed out. The bath was refilled and the indirectly-elicited twitches resumed. Two control responses were obtained for Acetylcholine.
Having set up the previously mentioned controls, 0.3ml of 1*10-6M Tubocurarine was added, whilst the twitches were still on. After the effects of Tubocurarine had come to equilibrium (approximately 10 minutes), the twitches were switched off and the response to Acetylcholine of afore mentioned concentration was tested. After the response was noted the twitches were switched on and the organ bath washed out.
The next step of the experiment once the preparation had recovered was to add 0.2ml of 1*10-7M Succinylcholine and the effects of the drug allowed to reach equilibrium at which point the drug was washed out. The application of Succinylcholine was repeated, however this time at the peak response Tubocurarine was added (same concentration as used before) and the effects noted.
The next step of the experiment was to add Tubocurarine to the bath and the response allowed to reach equilibrium. At this point Edrophonium was added and the effects noted. Having done this the organ bath was washed out and the preparation allowed to recover. Then Succinylcholine was added to the preparation and once its effects reached equilibrium, Edrophonium was added and the results noted.
             Questions.
1- From the experiments that were performed it can be said for certain that Tubocurarine is acting postjunctionally. The reason for this conclusion is as follows;.
After the addition of Tubocurarine to the organ bath, the effects of the twitches were greatly decreased until there was almost no response.
